tected with a drying tube. A solution of phosgene (55 ml, 12.5% in benzene) was then added with vigorous stirring. The reaction was cooled for an additional 10 min and was then kept at room temperature for 1 hr. The reaction mixture was cooled again, treated with 100-ml portions of 1 N HCl until free of amine, and then rinsed successively with 8% NaHCOs and water, dried, and evaporated, giving a thick oil which solidified in methanol. Electron-conformational study for the structure-hallucinogenic activity relationships of phenylalkylamines.
The structure-hallucinogenic activity relationships of a series of phenylethylamine and phenylisopropylamine derivatives have been investigated in the frameworks of electron-conformational method. The calculated geometry and electronic structure parameters accompanying to each atom and bond of each molecule in view were arranged as a matrix called electron-conformational matrix of contiguity (E...
متن کاملStructure-activity correlations for psychotomimetics. 1. Phenylalkylamines: electronic, volume, and hydrophobicity parameters.
CNDO/2 calculations have been performed on a series of alkyl, alkoxy, and alkylthio derivatives of phenethylamine and phenylisopropylamine. The results of these, of van der Waals volume calculations, and of Hansch type hydrophobicity calculations were correlated with psychotomimetic activity by chemometric methods. Eight parameters, involving seven chemical descriptors, were found to be highly ...
متن کاملArylalkylamine drugs of abuse: an overview of drug discrimination studies.
Abused arylalkylamines fall into two major categories: the indolealkylamines, and the phenylalkylamines: These agents can be further subclassified on the basis of chemical structure. Examples of these agents possess hallucinogenic, stimulant, and other actions. Drug-discrimination techniques have been used to classify and investigate this large family of agents. Such studies have allowed the fo...
متن کاملA QUANTUM CHEMICAL STUDY OF STRUCTURE -ACTIVITY RELATIONSHIPS OF DIHYDROPYRIDINE CALCIUM ANTAGONISTS
Quantum chemical (MNDo)caleulations have beenused toelucidate the molecular properties and structure - activity relationships of dihydropyridine (DHP) type calcium antagonists. There is a good corielation between the net atomic charges on various atoms of the 4 - phenyl ring of dihydropyridines and pharmacological activity. .Also, activity decreases with increasing free rotation of the phen...
متن کاملPhenylalkylamines with potential psychotherapeutic utility. 2. Nuclear substituted 2-amino-1-phenylbutanes.
A series of 2-amino-1-(4-substituted-2,5-dimethoxyphenyl)butanes (Table V) was prepared as analogues of (R)-2-amino-1-(2,5-dimethoxy-4-methylphenyl)butane (1a). 1-(2,5-Dimethoxyphenyl)-2-(N-phthalimido)butane (7) was utilized as a synthetic intermediate common to many of the target compounds. Animal data are presented indicating that most of these analogues have low hallucinogenic potential. Se...
